What is Capivasertib?

Capivasertib is an oral medication currently being investigated as a treatment for certain types of cancer, particularly HR-positive, HER2-negative breast cancer with PIK3CA mutations. It is often used in combination with other treatments such as aromatase inhibitors (e.g., letrozole) or other targeted therapies.

Capivasertib is part of a class of drugs known as AKT inhibitors, and it works by blocking a protein called AKT, which plays a critical role in the growth, survival, and spread of cancer cells.

How Does Capivasertib Work?

Capivasertib works by targeting and inhibiting AKT, a protein that helps regulate key cell processes such as growth and survival. When cancer cells have overactive AKT signaling, they can grow uncontrollably. By blocking AKT, capivasertib aims to slow or stop cancer cell growth, making it easier for other treatments (such as chemotherapy or hormonal therapy) to work effectively.

How is Capivasertib Taken?

  • Form: Capivasertib is taken as a tablet.

  • Elevated Liver Enzymes: Capivasertib can affect your liver function, leading to changes in liver enzymes. Your doctor may monitor your liver health through blood tests.

Serious Side Effects:

  • Liver Damage: Capivasertib can cause liver toxicity. Signs of liver problems include yellowing of the skin or eyes (jaundice), dark urine, or pain in the upper right side of your abdomen. Notify your doctor immediately if you experience any of these symptoms.
  • Infections: Capivasertib can affect your immune system and increase the risk of infections. Contact your healthcare provider if you have symptoms of an infection such as fever, chills, or sore throat.
  • Severe Diarrhea or Dehydration: Prolonged or severe diarrhea can lead to dehydration, so it’s important to stay hydrated. If diarrhea is persistent, seek medical advice.
  • Heart Problems: Though rare, capivasertib may cause heart problems such as irregular heart rhythms. Symptoms may include shortness of breath, dizziness, or chest pain. Let your doctor know if you experience any of these symptoms.

Precautions Before Taking Capivasertib

Before starting capivasertib, inform your healthcare provider if you:

  • Have Liver Problems: Since capivasertib can affect liver function, it’s important to inform your doctor if you have any history of liver disease.
  • Have Heart Problems: If you have a history of heart disease or irregular heart rhythms, let your doctor know, as capivasertib can affect the heart.

What to Avoid While Taking Capivasertib

  • Grapefruit and Grapefruit Juice: Avoid consuming grapefruit or grapefruit juice, as they may interfere with how capivasertib is processed in your body.
  • Live Vaccines: Do not receive live vaccines while taking capivasertib unless advised by your doctor. Live vaccines may not be safe during treatment.
  • Infections: Capivasertib can weaken your immune system. Avoid close contact with people who are sick and practice good hygiene, such as frequent handwashing.

Regular Monitoring

Your healthcare provider will monitor your health regularly while you are on capivasertib. This may include:

  • Blood Tests: Your doctor will perform blood tests to monitor your liver function, kidney function, blood counts, and blood sugar levels.
