FOLFOX Regimen Schedule

The FOLFOX regimen typically consists of the following drugs administered in 14-day cycles:

Leucovorin	400 mg/m²	2-hour IV infusion on Day 1
5-Fluorouracil	400 mg/m²	IV bolus on Day 1
5-Fluorouracil	2400 mg/m²	46-hour continuous IV infusion

Patients usually receive up to 12 cycles of FOLFOX, taking around 6 months in total

Efficacy

FOLFOX has been shown to be effective as adjuvant therapy for stage III colon cancer:

• The MOSAIC trial demonstrated improved disease-free survival and overall survival with FOLFOX compared to 5-FU/leucovorin alone
• At least 8 cycles of FOLFOX are needed to have an overall survival benefit, and 7 cycles for disease-free survival benefit
